Procedure:

Make miniprep and digest with EcoRI.

PCR Screening

Result:

Digest with EcoRI

PCR screening

Use forward primer and reverse primer from Biolabs.

Send sample 1 and 5 for sequencing.

EXP2

Purpose: PCR using Trec-combo template and Sample 1 in EXP1.

Procedure:

Taq: Lot 67-2038 PCR, 0.5ul.

PCR cycle:

1 min at 94 oC

30 cycles:
30 s at 94 oC
30 s at 55 oC
1 min at 72 oC

5 min at 72 oC
