Ligation of DNA fragment and pMiniT2

1.Contratc out with company to synthsize DNA fragments.

2.See the instruction of the related DNA fragments.

3.Use 10ul to disolve the Samples.

4.Transfer 1 ul to ligate with pMiniT2 Vector.

5.Formula:

a.1 ul diluted DNA fragments+3 ul H₂O

b.0.5 ul pMiniT2 vector

c.4ul Cloning Mix 1(buffer)

d.1 ul Cloning Mix 2(Ligase)

6.Ligate at RT for 15 min.

7.Then do transformation.
