No band in lane 3, expected size ~1.7 kb. Need to reamplify on Monday.

Cut out top band in lane 2, purified and digested
Kpn1 + Nco1 digestion of the vector will cut out MBP, so cut the vector (the largest fragment), see circled fragment in lane 3. Cut out insert and vector, purified and ligated for an hour for transformation
Colony Screening

Grew samples 1, 3, 5, 6, 7 and 8 in media (lanes 9, 11, 13, 14 and 16) overnight

Digested miniprep with enzymes to check if insert is still present
Samples 1, 2, 3 and 6 were positive (expected size: ~1.7 kb)
Submitted sample 3 (lane 9) for sequencinng.
