Laemmli SDS-PAGE buffer

10X electrode buffer stock
Tris base 30.3 g
Glycine 144.0 g
SDS 10.0 g

Dissolve in water to a total volume of 1 liter. Do not adjust the pH. Store at room temperature. To prepare 1X electrode buffer (0.025 M Tris, 0.192 M glycine, 0.1% SDS, pH 8.3), dilute 100 ml of 10X buffer stock with 900 ml of water.

Tricine SDS-PAGE buffer

10X electrode buffer stock
Tris base 12.1 g
Tricine 17.9 g
SDS 10.0 g


Dissolve in water to a total volume of 1 liter. Do not adjust the pH. Store at room temperature. To
prepare 1X electrode buffer (0.1 M Tris, 0.1 M Tricine, 0.1% SDS, pH 8.3), dilute 100 ml of 10X
buffer stock with 900 ml of water.

Laemmli SDS-PAGE buffer

Separating Gel Buffer, pH 8.8
1.5 M Tris-HCl:
51 g of Tris-HCl,  52.5 g Tris-Base

4 g SDS
Add water to final volume 500 mL, check pH, should be around 8.8

Stacking Gel Buffer, pH 6.8
1 M Tris-HCl
78.8 g of Tris-HCl in H2O,

2 g SDS
Final volume 500 mL, check pH, should be around 6.8, if not, adjust with HCl
