Grow condition:

Induce condition: 0.15 mM IPTG, room temperature o/n

Lysed with Buffer A without Imidazole. Load to 5 ml GE/Pharmacia Ni FF.

Elute from Ni column using linear imidazole 10 to 30 mM. Combine peak fractions, dilute with buffer to lower salt conc ~50 mM NaCl, load to 20 ml SP HP column (1.6 cm ID).  proteins do not bind to SP HP, load back to 10 ml Ni HP column, eluted as a single peak, analyse protein by SDS_PAGE. Looks like protein expression is rather low
