PKA alpha purification 2011 6 17

Use new protocol and Qiagen column (5 ml) PKA Purification

Run SDS-PAGE

10  ul Fraction 2 to 10, ---> lane 1 to 9 see notebook page 2930-2932;

combine fraction 2 to 6, use pbs/10% glycerol dialysis o/n;

before dialysis, od 280 is 0.410; afer dialysis, od 280 is 0.504;

estimate protein concentration is around 0.5mg/ml.

aliquot to 1.5ml tube, freeze in -80 degree (in the 0.1mg/ml pka, 10%glycerol box, use the red marker).

20 ul samples from fraction 1 through 9 (note book 002801, Ruili Su);

Combine faction 1 to 9, use PBS with20% glycerol to dialysis O/N;

Then adjust the protein concerntration until 1.5mg/ml by centrifuge.
