Adenovirus Amplification 293F

1. Grow 293F cells in 293 SFM II (with 4 mM l-glutamine) medium in suspension in a T-flask at 37 °C, 5% CO2.

2. Infect 3X10^5 cells in 293 SFM II medium with serotype 5 adenovirus (Ad5, with deletions in the E1 and E3 regions), at an MOI of 0.1 TU per cell. Replace culture medium every two days.

3. Harvest cells 8 days after infection.

Yield: 1.2 X 10^11 TU with 60 ml medium used.
