20 oligo first round Two PCR reactions, then Using outside primers second round PCR, fragement ~800bp

EcorI+Sal1 digest pET28b and PCR fragement, gel purified, ligation O.N at 16 degree.

8 clones screening( after ECoRI and Sal1 digested):

Picked clone 3 for sequencing, using T7 term as seq primer
