Lentivirus QC
Virus supernatant.
[edit]1. 2 days after transfection, take out 0.5 ml medium and add to 1 well of 293T cells growing on 12 well plate (50% confuluency).
2. Add 1000X polybrene so the final concentration is 1X
3. Incubate for 2 to 5 days and monitor cells for infection (GFP or RFP).
4. Check cells for fluorescence and bacterial contamination.
Purified virus
[edit]1. Add 1, 2.5, 5 and 10 ul of purified (concentrated) virus to 1 well of 293T cells growing on 12 well plate (50% confuluency).
2. Add 1000X polybrene so the final concentration is 1X
3. Incubate for 2 to 5 days and monitor cells for infection (GFP or RFP).
4. Check cells for fluorescence and bacterial contamination.
