Vaccinia DNA polymerase can be purified by.  A working stock of polymerase was prepared by diluting it to a concentration of 25 ng/mL in polymerase dilution buffer.

A C-terminal His6-tagged form of recombinant vaccinia single-strand DNA binding protein (I3)  E. coli strain BL21 and purified using nickel affinity columns as described by Tseng et al. (18). The purity was confirmed by SDS-PAGE, and the concentration determined using a Bradford assay. To produce a working stock, the I3 DNA binding protein was diluted to a concentration of 0.5 mg/mL in PBS.
