PHP-PEI conjugation optimization

1. Synthesis of PHP

1.1: 1.5g 1,3-diamino-2-propanol 16.6 mmole

+ 3 ml MeOH to dissolve

+ 3.63 g 1,3-dibromo-2-propanol, 16.6 mmole

1.2: 50 oC for 48 hrs

1.3: add 5 X5 ml MeOH to wash, spin, remove soluble material, collect pellet.

1.4: add 5 ml H2O to pellet to dissolve

1.5: add 30 ml EtOH to precipitate, spin, remove supernatant.

1.6: dry at 50 oC incubator.--->0.8 g polymer.

2. Synthesis of PHP-CBA-PEI18

TR7	50 mg	50 mg	25 mg
TR8	50 mg	10 mg	12.5mg
TR9	50 mg	10 mg	25 mg
TR10	50 mg	5 mg	12.5 mg
TR11	50 mg	5 mg	5 mg
TR12	50 mg	10 mg	50 mg
TR13	50 mg	10 mg	5 mg

adjust to 1ml with water:MeOH (9:1).

50 oC incubate o/n.
