The vector 433  pDB.His.GST was digested by NdeI/XhoI

Digest the PCR product, ligation, transformation.

There wasn't any colony growing on the plate.

Repeat again, same result as last one, no colony growing on the plate.

Redo again, there was one colony growing on the plate. Mini preparation, digestion.

Colony Screening:

There was a faint band around 300bp, but hard to see. Redigest again,

Still hard to see. Maybe it wasn't a positive plasmid. Or need other method to verify it is a positive plasmid.
