Protocol 1

[edit]

22.8 g NaH2PO4 (mw=120; 0.038M)  or 26.2g NaH2PO4(H2O) (mw=137.99; 0.038M) (Note: monobasic)
115g Na2HPO4 (mw=141.96; 0.162M) (Note: dibasic)
438.4g NaCl

Make it up

Add NaH2PO4 and Na2HPO4 and NaCl to 4500 ml dH2O.

Adjust pH to 7.4 by adding dilute (1N) HCl if necessary.

Bring the final volume to 5000 ml with water.

Dilute 1:10 for final concentration.

Protocol 2

[edit]

137 mM NaCl
2.7 mM KCl
100 mM Na2HPO4
2 mM KH2PO4

To make 10X PBS Buffer, dissolve the following reagents in 800 ml ultrapure water.
NaCl 80 g
KCl   2 g
Na2HPO4 14.4 g
KH2PO4 2.4 g

Adjust pH of PBS Buffer Solution to 7.4 with HCl.
Bring volume to 1 liter, autoclave or sterilize by filtration.

Protocol 3

[edit]

A 10 liter stock of 10x PBS can be prepared by dissolving 800 g NaCl, 20 g KCl, 144 g
Na2HPO4 · 2H2O and 24 g KH2PO4 in 8 L of distilled water, and topping up to 10 L. The
pH is ~6.8, but when diluted to 1x PBS it should change to 7.4. When making buffer
solutions, it is good practice to always measure the pH directly using a pH meter. If
necessary, pH can be adjusted using hydrochloric acid or sodium hydroxide.
On dilution, the resultant 1x PBS should have a final concentration of 137 mM NaCl, 10
mM Phosphate, 2.7 mM KCl, and a pH of 7.4.
