Yeast Genomic DNA Extraction

Pick a single colony Saccharomyces cerevisiae from a YPD plate

Suspend in 100 μl of 200 mM LiOAc 1% SDS solution

Incubate at 70°C for 15 minutes.

Add 300 μl of 96% ethanol

Spin 15, 000 g for 3 minutes

Dissolve DNA in 100 μl of TE

Spin down cell debris for 15 000 g; 1 minute

Use 1 μl of the supernatant  for PCR
